Ribonuclease U2, Recombinant - Manual
Ribonuclease U2 is an endonuclease that hydrolyzes the phosphodiester bonds of 3’-linked ribonucleotides, resulting in nucleoside 3’-phosphates and 3’-phosphooligonucleotides ending in Ap or Gp with 2’, 3’-cyclic intermediates. It is purine selective with a preference for adenosine and has the following order of cleavage specificity (A>G>>C>U). Because of this, it is useful in RNA sequencing and RNA modification and mapping applications. Optimal activity is observed at a pH of 4.5.
Ustilago sphaerogena produces ribonuclease U2 recombinantly in yeast Komagataella phaffii (previously P. pastoris) to decrease the amounts of contaminant nucleases and proteases and eliminate potential pathogens associated with animal-based materials.
